Cover Story (view full-size image): Among all the human barriers, the blood–brain barrier (BBB) maintains the physiological homeostasis of the brain. BBB’s continuous non-fenestrated capillaries system allows precise homeostasis regulation and protection. From a pharmaceutical point of view, it notably complicates the treatment of brain disorders since it impairs chemical drug and biopharmaceutical delivery into the brain reducing their therapeutic efficacy and/or increasing their unwanted bioaccumulation in the surrounding healthy tissues. Nanotechnological innovations provide advanced biomaterials and customized engineering and functionalization methods for brain-targeted drug delivery. Lipid nanocarriers, both synthetic and cell-derived ones, are considered effective brain delivery systems due to their reasonable biocompatibility and ability to cross the BBB.View this paper
