Cover Story (view full-size image): Pseudotyping expands the tropism of lentiviral vectors (LVs), enhancing their utility. While vesicular stomatitis virus G (VSV-G) single-pseudotyped LVs (V-LV) are common, dual-pseudotyping is rare due to its complexity. In this study, we investigated dual-pseudotyped LVs with both VSV-G and Sendai virus hemagglutinin-neuraminidase (SeV-HN) glycoproteins, termed V/HN-LV. We demonstrated the significantly improved transduction efficiency of V/HN-LV in mouse, monkey, and human cells, including primary human hematopoietic stem cells. SeV-HN recognizes sialylated receptors on the host cell membrane and cleaves sialic acid from VSV-G. These synergistic actions increase infectivity and confer the dual-tropic competence of V/HN-LV, suggesting its potential applications from basic science to ex vivo human gene therapy.
