PHYSICAL ACTIVITY REDUCES BRAIN STRESS ACTIVITY TO LOWER RISK OF CVD https://lnkd.in/eqb4aKaw April 17, 2024   New research published in the Journal of the American College of Cardiology led by Massachusetts General Hospital, a founding member of the Mass General Brigham Healthcare System indicates that physical activity lowers the risk of cardiovascular disease, in part, by reducing stress-related signaling in the brain, finding that those with stress-related conditions such as depression experienced the most cardiovascular benefits from physical activity. The researchers analyzed medical records and other information from 50,359 participants enrolled in the Mass General Brigham Biobank who completed a physical activity survey, and a subset of 774 participants who underwent brain imaging, tests, and measurements of stress-related brain activity to assess the mechanisms underlying the psychological and cardiovascular disease (CVD) benefits of physical activity.  The analysis revealed that over a median follow-up of ten years, 12.9% of the participants developed CVD, and those who met physical activity recommendations had a 23% lower risk of developing CVD compared to those who did not meet the recommendations.  After accounting for a wide range of lifestyle variables and other risk factors, those with higher levels of physical activity were found to have lower levels of stress-related brain activity. Reductions in stress-associated brain activity were driven by gains in function in the prefrontal cortex which is involved in executive function and is known to restrain stress centers of the brain.  The reductions in stress-related brain signaling account for physical activity’s cardiovascular benefit. Further analysis revealed that in a cohort of 50,359 participants the cardiovascular benefits of exercise were substantially greater among those who would be expected to have higher stress-related brain activity, such as those with depression. "Physical activity was roughly twice as effective in lowering cardiovascular disease risk among those with depression. Effects on the brain's stress-related activity may explain this novel observation," said Ahmed Tawakol, MD, who is the senior author of the study, an investigator and cardiologist in the Cardiovascular Imaging Research Center at Massachusetts General Hospital. "Prospective studies are needed to identify potential mediators and to prove causality. In the meantime, clinicians could convey to patients that physical activity may have important brain effects, which may impart greater cardiovascular benefits among individuals with stress-related syndromes such as depression." 💠REPORT | 2024 #AACR Annual Meeting From the Tisch Cancer Institute at Mount Sinai in New York, Professor Sundar Jagannath presented "Linvoseltamab Induces Deep Responses in Advanced Relapsed/Refractory Multiple Myeloma." According to the assessment by an independent review committee at 11.1 months, the objective response rate (ORR) for BCMA-targeted T-cell-engaging #bispecific #antibody 200 mg once weekly dosing was 71% (n=117), with 41% of patients achieving stringent complete responses (sCR), 46% achieving complete responses (CR) or better, and 62% achieving very good partial responses (VGPR; PR) or better. According to data from the Phase I/II LINKER-MM1 study (NCT03761108) presented at the meeting, Linvoseltamab demonstrated high response rates and acceptable safety characteristics in patients with relapsed/refractory multiple myeloma (including refractory subgroups). "Linvoseltamab has shown high efficacy in patients with advanced relapsed/refractory multiple myeloma, including those in predefined high-risk subgroups," Professor Sundar Jagannath said during the presentation. "The safety profile of linvoseltamab is acceptable, with the most common treatment-emergent adverse events [TEAEs] being cytokine release syndrome [CRS], neutropenia, and anemia." Other data showed high response rates in predefined subgroups. Jagannath stated, "You can see response rates of around 70% in patients aged 75 years or older, African Americans, patients with high-risk cytogenetics, or ISS III stage patients. However, patients with extramedullary disease or bone marrow plasma cells ≥50% or soluble BCMA ≥400ng/mL had response rates of 48-56%." For heavily pretreated multiple myeloma patients, bispecific antibodies are an effective option, with ORRs ranging from 61% to 63%. The fully human BCMAxCD3 bispecific antibody linvoseltamab requires patients to be hospitalized twice for one day each time, allowing for monthly dosing, and has been shown to induce deep responses in patients. Highlights of the LBR indicate significant progress in the field of bispecific antibodies for cancer treatment, with several heavyweight studies selected for oral presentations and posters at the conference. I listened to eight pharmaceutical companies report their groundbreaking progress in exploring bispecific drugs. For example, Johnson & Johnson presented "JNJ-87890387, a novel ENPP3 bispecific T-cell redirector (ENPP3xCD3) with tumor selectivity through targeting apical surface antigens," reporting recent significant progress in tumor treatment through bispecific approaches. Additionally, the National Cancer Institute of the United States presented "Unveiling the potential of bispecific antibodies: A comprehensive analysis of efficiency, toxicity, and repurposing in cancer treatment," revealing the enormous potential of bispecific antibodies and providing a detailed explanation of their comprehensive effects on efficacy, toxicity, and reuse in cancer treatment.

This study "aimed to conduct a pilot study to measure Interoceptive bodily awareness (IBA), with the Multidimensional Assessment of Interoceptive Awareness (MAIA), in patients seeking OMT for pain, and to test if OMT exposure may be associated with higher IBA as measured by the MAIA." Exciting new research from the Center for Healthy Aging and investigators Waylon Hastings and Idan Shalev on the effects of diet (specifically, caloric restriction) on telomere loss. This work also involved Biobehavioral Health graduate student Qiaofeng Ye.
