Cover Story (view full-size image): Accurate diagnosis of Alzheimer’s disease (AD) and Frontotemporal dementia (FTD) represents a health issue due to the absence of disease traits. Biomarkers have gained a pivotal role in clinical trials, as they accurately reflect the underlying pathology in pre- or oligosymptomatic stages of different dementias. Cerebrospinal fluid (CSF) SIMOA biomarkers exhibit increased levels in AD and FTD samples compared to controls. Moreover, novel statistical models based on a combination of different biomarkers and demographics and clinical patients’ characteristics classify the dimensional clinical datasets into confident disease clusters, highlighting the need for more representative samples, variables, and measurements for successful and more efficient cohort clustering.
