Cover Story (view full-size image): Sublingual vaccines stimulate both mucosal and systemic immune responses, producing antigen-specific s-IgA in mucosae and both IgG and IgA in blood. This protects against viruses in the respiratory tract, like SARS-CoV-2 or influenza. The sublingual vaccine can be self-administered without a needle. Previously, we developed a sublingual vaccine formulated with SARS-CoV-2-RBD antigen and Poly (I:C) in nonhuman primates (NHPs) (doi.org/10.1093/biomethods/bpad017, doi:10.26502/ami.936500113). In the current study, we reported the efficacy and safety of a sublingual vaccine with Influenza HA antigen and Poly (I:C) adjuvant in NHPs. This study also revealed previously unknown mechanisms underlying immune responses mediated by the sublingual vaccine, inducing a “Yin/Yang"-like balance state of immune-enhancing and -suppressive responses.
