Zhao group has developed a new reagent for the acid-amine coupling for the peptide bond synthesis. Allenone mediated peptide bond formation got its advantage for the racemization and epimerization free coupling process. They have given a library of examples where the coupling between the different amino acids and fragments went very smoothly via the formation of a vinyl ester. The group had synthesized an 'onco'peptide (Carfilzomib) with the same process and got a reasonably high purity and yield compared to the previously reported process. Allenone mediated vinyl esters of different amino acids (simply called as active ester of the inactive amino acid)can be used in SPPS process and which gives a reasonably high purity compared to the well known 'aryl ester amino acid' pathway. Chains of fewer than ten or fifteen amino acids are called oligopeptides, and include dipeptides, tripeptides, and tetrapeptides. A polypeptide is a longer, continuous, unbranched peptide chain.[3] Hence, peptides fall under the broad chemical classes of biological polymers and oligomers, alongside nucleic acids, oligosaccharides, polysaccharides, and others. A polypeptide that contains more than approximately fifty amino acids is known as a protein.[3][4][5] Proteins consist of one or more polypeptides arranged in a biologically functional way, often bound to ligands such as coenzymes and cofactors, or to another protein or other macromolecule such as DNA or RNA, or to complex macromolecular assemblies. Amino acids that have been incorporated into peptides are termed residues. A water molecule is released during formation of each amide bond. All peptides except cyclic peptides have an N-terminal (amine group) and C-terminal (carboxyl group) residue at the end of the peptide. A CDR region, also called Complementarity Determining Region, is a sequence of amino acids that folds into loops capable of binding to an antigenic amino acid sequence, also known as an epitope. Both the light and heavy antibody chains are made of variable (V) and constant (C) regions; each variable region contains three CDR loops that form the majority of the antigen-binding site. Outside of the CDR loops, framework sequences stabilize the loop structure. Check out the figure below! Given the substantial variability in CDRs, pinpointing the exact amino acid sequence can be challenging due to their novel and unique nature. Each CDR represents a distinct protein sequence, setting them apart from one another. Cannabidiolic acid (CBDa), a precursor to CBD, represents one of the primary raw cannabinoids, alongside THCa and CBCa. These compounds play a crucial role in modulating the endocannabinoid system (ECS), fostering balance and well-being within the body. ⚛️ Delving into the chemistry, CBDa consists of 22 carbon atoms, 30 hydrogen atoms, and four oxygen atoms, forming the foundation of its molecular structure. 🚀 Via decarboxylation, or exposure to low heat, CBDa converts to its famous metabolite, CBD. A metabolite is the end product of a chemical process; in this case, decarboxylation. 🌿 CBDa lacks psychoactive properties, meaning it does not get you “high”.
