The Human Pangenome In 2003, biologists created the first ever human genome sequence. The 3 billion DNA letter sequence, called the reference genome, was mostly made up of DNA donated from people in the city of Buffalo, New York. So far, when clinicians and researchers study an individual’s genome, they compare it to the reference genome to identify differences. But can you compare all of humanity to one genome? No, because one reference genome does not convey the genomic diversity of the human species. We need many reference genomes--a pangenome. This monumental undertaking is already taking place and is poised to redefine the future of genomic research and human health. The Cleft Collective was set up in 2012, with the goal of collecting genetic samples and demographic data from children born with cleft and their parents. Over 10,000 mums, dads, and babies have taken part - making it one of the largest bibanks in the world! Using this enormous dataset, researchers are digging deep into what causes cleft, what the best treatments are, and how a cleft affects development. The Cleft Collective is now being funded and managed by The Underwood Trust, but the data we have collected from volunteers across the UK is helping to make scar free healing a reality. The NIH is conducting planning activities to inform a potential Common Fund research program to catalyze the development, standardization, validation, and use of new methods and approaches that will more accurately model human biology, known as New Approach Methodologies (NAMs). We anticipate these new methods and approaches would complement, or in some cases, replace traditional animal models, transforming the way we do basic, translational, and clinical sciences. As part of the planning activities, the Common Fund will be hosting public listening sessions bringing together key representatives from multiple sectors to gain insight into current opportunities and roadblocks in NAM development unique to their fields. The next session will focus on learning more about communicating 3Rs (Replace, Reduce, Refine) considerations and highlighting public health concerns that could be addressed by NAMS from the non-government organization community. What would they say? 📣 I think that no scientist or geneticist has not heard of the famous revelation made by #Watson and #Crick in the Eagle Pub (yeah, that's right, a pub 😁) that they had discovered the #secretoflife. 🎖 Their discoveries, along with #Wilkins and #Franklin's, were formally presented scientifically and published in #Nature in April 1953. 🔎 Our comprehension of #DNA has rapidly grown over the last few decades, giving us unprecedented insight into the #genome. Naturally, we want to add more actionability to the data the more we find. #Genomecuration is one of the hottest subjects and objectives in science today. 📚 Given that Watson (1928–) and Crick (1916-2004)'s work forms the #foundation for everything we know about DNA today, I couldn't help but wonder what they would think of this aim, particularly if we highlight a timetable of roughly two years. 💊 As you are undoubtedly aware, Watson was the second person to #publicly release his entire genome online, with the intention of fostering the emergence of a #personalizedmedicine era that includes disease identification and prevention as well as the development of #customizedmedicaltreatments. 🔬 Whatever his #controversial remarks over the years (which are not the subject of this post), I sincerely hope he gets to see the #outcomes in the next two years and its #implementation in the best possible light. 🎊 My humble contribution to this curating project, makes me very proud and thankful. I can easily picture myself celebrating with friends and coworkers when a goal is accomplished—maybe not in a pub, but in some Bosnian "kafana."
