Shocking 😳 this isn’t good news at all. An example: using horses to test anti-arthritis drugs for humans. We have human relevant methods available and millions of arthritis patients data to use. This animal screening platform is totally unnecessary. “Today, a startup called Gordian Biotechnology debuted a technology that could make this process better for both animals and people. The San Francisco-based company has developed a new animal screening platform which allows multiple gene therapies to be tested at the same time with just one animal. Instead of the gene treatment being given to the animal and affecting an entire area of its body, Gordian’s innovation enables it to test a drug inside of a single cell. That means one mouse could potentially support the evaluation of hundreds of possible new gene therapies in a way that’s faster and impacts fewer animals with less risk of harm to them” https://lnkd.in/ejwKU9jC

To better predict human outcomes in the drug discovery process, animal models must model the patients we hope to cure. So for diseases of aging, it makes perfect sense to study aged animals. But until now, the number of animals needed made this practically impossible. Even if you could afford and find thousands of horses with naturally occurring osteoarthritis, you couldn’t afford to keep them. Today, Gordian unveiled notable results from its in vivo screening platform that uses recent innovations in single-cell sequencing and gene therapy to test hundreds of therapies simultaneously in one sick animal. And just like that, there’s an entirely different drug discovery construct for complex age-related diseases. Gene silencing is a powerful tool utilized for identification of gene function and analysis in plants, animals, and fungi. Here, we report the silencing of superoxide dismutase (bcsod1) in Botrytis cinerea through sense and antisense-mediated silencing mechanisms. Because superoxide dismutase (SOD) is a virulence factor, transformants were tested for phenotypic silencing in vitro and reduction in pathogenicity in planta. Plate-based assays with and without paraquat were performed to screen initial silencing efficiency, and a subset of transformants was used for in planta studies of virulence. Transformants exhibiting strongly decreased transcripts levels were recovered with both constructs but none of those exhibited a reduction in virulence in planta. Our investigations may help optimize a high-throughput gene silencing system useful for identifying potential gene targets for future fungal control.

Heritable transgene-free genome editing in plants by grafting of wild-type shoots to transgenic donor rootstocks The key hypothesis of this research may have been that graft-mediated transfer of CRISPR-Cas9 components using tRNA-like motifs could enable efficient gene editing in plant systems without the need for extensive outcrossing or transgene elimination. The study demonstrated the fusions of CRISPR-Cas9 and guide RNA transcripts with tRNA-like motifs facilitated graft-mediated RNA transfer in plants. This approach enabled heritable gene editing in wild-type Arabidopsis thaliana and Brassica rapa without the necessity of transgene elimination or viral vectors. The graft-mobile gene editing system allowed the production of transgene-free offspring in a single generation, bypassing the lengthy process of culture recovery and selection. The results suggested a promising method for efficient and transgene-free crop improvement, with potential applications in other crops. https://lnkd.in/g84ABWdf
