Read this article in Nature 📚🔬about the crucial role of the cGAS-STING pathway in age-related inflammation and disease. It's not our work, but important to share it. The research found that low-grade inflammation, a common feature of old age, is significantly driven by the cGAS-STING signaling pathway, a vital mechanism for immune response to DNA. 🧬 The team demonstrated that blocking STING could mitigate the inflammation associated with aging in both human cells and mice tissues. 🐁👩🔬 Remarkably, they also revealed that activation of STING in the brain could lead to cognitive decline and neurodegeneration. This underlines the cGAS-STING pathway's potential role in neurodegenerative diseases. 🧠💡 The exciting implication? Targeting this pathway could be a future strategy to combat aging-related inflammation and neurodegeneration. 🚶♀️ A healthy diet and exercise obviously have lots of benefits, including for people living with cancer. Researchers at the Yale University School of Medicine have been looking at whether a targeted diet and exercise intervention could help women undergoing chemotherapy for breast cancer. Speaking to the Yale School of Medicine, lead author Tara Sanft said that they wanted to explore what could help people with breast cancer have an “easier time completing more of their chemotherapy.” Published in the Journal of Clinical Oncology, this randomised control trial tested an intervention that  involved counselling, as well as following healthy eating and physical activity guidelines (which included brisk walking), over 3 months. Chemotherapy completion rates were similar for women randomly assigned to intervention compared to usual care. Alongside improvements in diet quality, physical activity levels, the intervention led to reported improvements in pathological complete response rates compared with usual care (UC). The authors suggest that this could be due to, “the effect of exercise and nutrition on pCR may be via different mechanistic pathways than chemotherapy completion such as immune, inflammatory, and metabolic pathways.” The researchers conclude that these findings could provide oncologists with a non-pharmacological intervention that may have  potential to improve outcomes. More robust research is needed to expand on these findings. DISTINGUISHING FEATURES OF LONG COVID IDENTIFIED THROUGH IMMUNE PROFILING Very relevant Mount Sinai-Yale Long COVID (MY-LC) paper published by Nature (Nature Portfolio). In this publication, more than 270 individuals with or without #longcovid (LC) were enrolled in a cross-sectional study that included multi-dimensional immune phenotyping and unbiased machine learning methods to identify biological features associated with LC. Marked differences were noted in circulating myeloid and lymphocyte populations relative to matched controls, as well as evidence of exaggerated humoral responses directed against #sarscov2 among participants with LC. These and other findings discussed in this article may help guide future studies into the pathobiology of LC and aid in developing relevant biomarkers. https://lnkd.in/eh42hcdP

Exciting new research by Dr Soyon Hong (UK DRI at UCL) has revealed fresh insight into the protective role of microglia, the brain’s resident immune cells, in early Alzheimer’s disease. Microglia are triggered to engulf, or eat, synapses – the connections between neurons – in the early stages of Alzheimer’s disease. This process, known as phagocytosis, leads to the loss of synapses, and this correlates strongly with cognitive decline. However, it is not yet known whether specific synapses are targeted, what the triggers are, or why the process occurs. The new study uncovered the mechanism by which microglia identify which synapses to engulf, and showed that microglia are acting in a protective capacity to reduce hyperactivity in synapses. Dr Hong explained: "Our results suggest that microglia are carrying out a protective mechanism, to help reduce hyperactivity in neurons in early Alzheimer’s disease and maintain a state of homeostasis in the cells. This study provides useful insight into how microglial function could be targeted in disease." Read the full story ⬇

Indeed, Abeta is an antimicrobial peptide. It was intriguing to observe its effects firsthand in a 3D matrigel system containing cells expressing human amyloid precursor protein (APP). When introduced to the system, Abeta actively coated E. coli bacteria, visible through time-lapse video. As the bacteria moved through the matrigel, the Abeta progressively slowed them down to a complete stop, effectively neutralizing them. This observation underscores the potent antimicrobial properties of Abeta within complex biological environments. #abeta #alzheimers #neuroscience #neurology
