  • Detection of a Foreign Protein in Milk Using Surface-Enhanced Raman Spectroscopy Coupled with Antibody-Modified Silver Dendrites

    Analytical Chemistry

Herein we developed a rapid and simple method which used surface-enhanced Raman spectroscopy (SERS) coupled with antibody-modified silver dendrites to detect ovalbumin (OVA), the egg white protein, introduced into whole milk. OVA was first captured out of milk by use of antibody-modified silver dendrites and then directly measured on the silver dendrites by Raman spectroscopy. Results show that this method is capable of detecting OVA at 0.1 μg/mL in phosphate buffered saline (PBS) and 5 μg/mL…

This method has the potential for wide use in areas such as allergenic protein detection and bioterrorism agent detection in complex matrixes.

  • Characterization of Amorphous and Microcrystalline Silicon Using Raman Spectroscopy

    Spectroscopy

Silicon deposited on glass or silicon carbide is widely used in manufacturing photovoltaic cells. Both the proportion and distribution of amorphous and crystalline silicon are critical for performance and are therefore important to monitor. Raman spectroscopy is an ideal technique for this application, as the two forms generate readily distinguishable spectra that lend themselves to simple quantification methods using Beer's Law. Mapping generates chemical images with detailed information on the spatial distribution of the crystalline and amorphous forms. Since excess excitation laser power can be demonstrated to convert amorphous into crystalline silicon, care must be taken to limit the amount of power used. The Thermo Scientific DXR Raman microscope, which is equipped with a laser power regulator, is ideal for this application, particularly if the method has to be replicated from instrument to instrument at multiple manufacturing plants.

  • Applications of low-resolution Raman spectroscopy (LRRS)

Internal Standardization and Calibration Architectures for Chemical Sensors, Proceedings of SPIE vol 3856

LRRS has been shown to have the potential to make Raman spectroscopy as practical and as widely used as IR spectroscopy. The advantages LRRS brings to Raman spectrometry are its order of magnitude lower price, size, and weight from that of laboratory grade Raman spectrometers. These allow the implementation of the Raman approach with all of its advantages in a small, lightweight, portable, and affordable instrument. The disadvantages of LRRS is a small degradation in performance from that of…

The disadvantages of LRRS is a small degradation in performance from that of laboratory grade Raman spectrometers. It is shown in this paper that for one class of applications, measuring the concentrations of analytes in a solvent, the degradation in performance is insignificant. This class of application includes monitoring monomers and polymer concentrations during polymerization and the chemical constituents during crystallization processes.

  • Stability and Surface Uniformity of Selected Thiol Coated SERS Surfaces

    Applied Spectroscopy

The use of surface-enhanced Raman scattering (SERS) as an analytical technique continues to grow, but questions about its viability remain. This paper addresses the concern about the long-term stability of the surfaces used with the SERS technique. We have studied the long-term stability of Ag surfaces coated with three different thiol compounds. For this study, we have used 1-propanethiol, 1-dodecanethiol, and p -cresolthiol to coat the Ag surfaces. These surfaces showed long-term stabilities…

These surfaces showed long-term stabilities of over a month with minimal surface degradation. In order to mimic a realistic application of SERS, we stored the coated surfaces in a solution of water. Sodium dodecylsulfate (SDS) was added to maintain consistent surface wetting conditions so that reproducible results were obtainable on a day-to-day basis. Spatial heterogeneity of the surfaces and coating was analyzed with a fiber-optic Raman system. The spatial heterogeneity of the surfaces explains the day-to-day variations observed in the detection when the same portion of the surface used for analysis varies from day to day.
