A team from Josephs's lab at the University of North Carolina published a study (DOI: 10.1002/smll.202403496 ) of an in vitro method, termed CCR (compartmentalized CRISPR reactions) for screening large numbers of gRNAs for both CRISPR effector activity and specificity by performing the complete CRISPR reactions within individual water-in-oil microemulsions. CCR could help streamline the gRNA validation processes for biomedical research applications, especially ones with high specificity. That anonymous reader was Mu Yang, an assistant professor of neurobiology at Columbia University, in New York City, who started emailing the journal about problematic papers in January 2023. I'm excited to share our latest review article, "Advancing clinical understanding of surface electromyography biofeedback: bridging research, teaching, and commercial applications." Expanding the use of surface electromyography-biofeedback (EMG-BF) devices in different therapeutic settings highlights the gradually evolving role of visualizing muscle activity in the rehabilitation process. This review evaluates their concepts, uses, and trends, combining evidence-based research. ⚡ This review dissects the anatomy of EMG-BF systems, emphasizing their transformative integration with machine-learning (ML) and deep-learning (DL) paradigms. Advances such as the application of sophisticated DL architectures for high-density EMG data interpretation, optimization techniques for heightened DL model performance, and the fusion of EMG with electroencephalogram (EEG) signals have been spotlighted for enhancing biomechanical analyses in rehabilitation. The literature survey also categorizes EMG-BF devices based on functionality and clinical usage, supported by insights from commercial sectors. * Electromyographic biofeedback (EMG-BF) has a significant role in providing real-time feedback for rehabilitation and enabling detailed analysis of muscle activation patterns, which is crucial for diagnosing and treating neuromuscular disorders. * EMG-BF devices come in three types: PC-based, handheld, and mobile/tablet-based. They vary in the number of channels and reliability based on scientific research. EMG-BF has multiple clinical applications, including mental and physical rehabilitation, facial expression detection, and stress analysis. * The SENIAM and ISEK promote standardized practices in sEMG through educational efforts and consensus papers, essential for ensuring reliable research and clinical applications. These efforts also aim to dispel misunderstandings about the scientific underpinnings of sEMG in physiotherapy. * The review paper also highlighted the importance of good training on the clinical application of EMG and how to perform EMG biofeedback exercises. Universities and global initiatives are enhancing sEMG education and its integration into clinical practice. This addresses training gaps and promotes effective utilization of sEMG technology to enhance patient care. * The AI applications on EMG biofeedback have been addressed and shown how machine and deep learning change the concept of EMG Biofeedback. Until relatively recently, Parkinson's Disease (PD) has primarily been considered a motor disorder, with its dominant clinical features the result of dopaminergic neuron degeneration. Although dopamine cell loss clearly has a major role in PD symptomatology and has been the basis for the development of dopamine-replacement symptomatic therapies, this focus on motor deficits has relegated non-motor problems to a secondary role. As a result, valuable insights on disease pathogenesis, clinical evolution and progression, and novel therapies have been missed or delayed 2. Imaging studies have showed that PD patients with depression exhibit reductions in dopaminergic and noradrenergic innervation in the locus coeruleus, thalamus and limbic brain regions and increases in neuronal loss and gliosis in the locus coeruleus (82,102,103) 3. Interestingly, antidepressant drugs that affect noradrenaline reuptake, namely, tricyclic antidepressants like nortriptyline, may be more effective than other drugs in the treatment of PD‑related depression (104) 4. Increased depression-like behaviour has been observed using the forced swim and sucrose preference tests in 6‑OHDA-lesioned and MPTP-treated rodents, and rotenone infusion into the substantia nigra in rats was associated with altered dopaminergic and serotoninergic transmission (89,105). Happy to share this important and inseideful information which is so valuable for the future developments and research in Neuroscience and Oncology: "This study investigates brain network modifications related to tumor grade and location using resting-state functional magnetic resonance imaging and graph theory. We demonstrated that low-grade gliomas (LGG) lead to increased efficiency of the surrounding functional network, while high-grade gliomas (HGG) seem to disrupt brain connectivity in remote areas. Tumor location appears to influence the pattern of reorganization, including the recruitment of the contralateral hemisphere. Overall, LGG may show more favorable connectivity changes than HGG. Cellular reprogramming by overexpression of Oct3/4, Klf4, Sox2, and cMyc (OKSM) transcription factors and rejuvenating effects: Background: With the elderly population projected to double by 2050, there is an urgent need to address the increasing prevalence of age-related debilitating diseases and ultimately minimize discrepancies between the rising lifespan and stagnant health span. Cellular reprogramming by overexpression of Oct3/4, Klf4, Sox2, and cMyc (OKSM) transcription factors is gaining attention in this context thanks to demonstrated rejuvenating effects in human cell cultures and live mice, many of which can be uncoupled from dedifferentiation and loss of cell identity. Summary: Here, we review current evidence of the impact of cell reprogramming on established aging hallmarks and the underlying mechanisms that mediate these effects. We also provide a critical assessment of the challenges in translating these findings and, overall, cell reprogramming technologies into clinically translatable antiaging interventions. Key Messages: Cellular reprogramming has the potential to reverse at least partially some key hallmarks of aging. However, further research is necessary to determine the biological significance and duration of such changes and to ensure the safety of cell reprogramming as a rejuvenation approach. A neat approach for killing EBV-associated tumour cells. As with any genetic-based therapy though, a challenge will be delivering the nucleic acid to sufficient cells for a therapeutic benefit... Synthetic BZLF1-targeted transcriptional activator for efficient lytic induction therapy against EBV-associated epithelial cancers. Abstract The unique virus-cell interaction in Epstein-Barr virus (EBV)-associated malignancies implies targeting the viral latent-lytic switch is a promising therapeutic strategy. However, the lack of specific and efficient therapeutic agents to induce lytic cycle in these cancers is a major challenge facing clinical implementation. We develop a synthetic transcriptional activator that specifically activates endogenous BZLF1 and efficiently induces lytic reactivation in EBV-positive cancer cells. A lipid nanoparticle encapsulating nucleoside-modified mRNA which encodes a BZLF1-specific transcriptional activator (mTZ3-LNP) is synthesized for EBV-targeted therapy. Compared with conventional chemical inducers, mTZ3-LNP more efficiently activates EBV lytic gene expression in EBV-associated epithelial cancers. Here we show the potency and safety of treatment with mTZ3-LNP to suppress tumor growth in EBV-positive cancer models. The combination of mTZ3-LNP and ganciclovir yields highly selective cytotoxic effects of mRNA-based lytic induction therapy against EBV-positive tumor cells, indicating the potential of mRNA nanomedicine in the treatment of EBV-associated epithelial cancers.
