#Pfizer reported results from a phase 3 trial, #CIFFREO, evaluating an investigational recombinant adeno-associated virus #genetherapy known as fordadistrogene movaparvovec among ambulatory boys ages 4 to 7 years with #Duchenne muscular dystrophy. CIFFREO failed to hit its primary goal of improving #motorfunction when compared to placebo at one year of treatment, as assessed by a movement scale known as the #NorthStarAmbulatoryAssessment. Key secondary endpoints, such as 10-meter run/walk velocity and #timetorise, also failed to demonstrate a significant difference between fordadistrogene movaparvovec and placebo. These results come a little more than a month after Pfizer reported the death of a young boy participating in a related phase 2 trial called DAYLIGHT, which included boys with DMD ages 2 to 3 years. The boy had received the gene therapy in early 2023 and died from cardiac arrest on May 3 of this year. It will be interesting to review more detailed results from both of these studies to better understand the reasons why the trial failed and more importantly, whether the death of the young boy was related to the treatment. Takeda inks sweet $1.2B+ deal with Kumquat for immuno-oncology program https://lnkd.in/eTtPuV3X Takeda is offering Kumquat Biosciences the chance to make more than $1.2 billion biobucks in a new exclusive pact that centers around an unnamed immuno-oncology small molecule inhibitor. Kumquat, after running phase 1 trial activities, can grant Takeda global licensing rights to develop and commercialize the asset, according to agreement terms shared April 16. Subject to Kumquat's option, Big Pharma Takeda will then take on responsibility for all development and commercialization activities following phase 1. The selected program can be developed as a monotherapy, combo therapy or both, according to the April 16 release. In exchange, Kumquat is set to make up to $130 million in near-term payments, with the additional chance to grab more than $1.2 billion in future clinical, regulatory and commercial milestones, plus tiered royalties on top of that.

#News: #Immunology is AbbVie’s world, everyone else just lives in it. That is the takeaway from ZOOMRX’s analysis of the perceptions of healthcare professionals (#HCPs), who overwhelmingly see #AbbVie as the top dog in a field targeted by rivals such as Pfizer and Johnson & Johnson. ZoomRx asked HCPs to rank immunology drug developers in terms of attributes such as #innovation and #patientcentricity. Most HCPs ranked AbbVie as one of the top three companies in each category, causing it to score 100 on ZoomRx’s perception scale. Pfizer was a distant second with a score of 70, followed by J&J with a score of 52. AbbVie was a dominant force in immunology throughout the #Humira era. Novel and #biosimilar rivals are starting to chip away at the megablockbuster, reducing its sales to $14.4 billion last year, but AbbVie has managed to stay at the top of the HCP rankings. On April 30th, The FDA granted full approval to #Pfizer and #Genmab’s antibody-drug conjugate Tivdak (tisotumab vedotin-tftv) for the treatment of recurrent or #metastatic #cervicalcancer that had progressed on or after #chemotherapy. Originally developed under a partnership between Seagen and Genmab, Tivdak is an antibody-drug conjugate (ADC) that targets the cell surface tissue factor protein, which is an important part of the blood coagulation cascade. Its toxic payload is monomethyl auristatin E, a microtubule-disrupting agent that triggers cell death. Pfizer took over Seagen’s half of the deal when it bought the cancer-focused biotech in March 2023 for $43 billion. J&J Buys Proteologix for $850M, Bispecific Antibodies for Atopic Dermatitis Johnson & Johnson is expanding its dermatology portfolio with an $850 million cash deal. The pharma giant announced Thursday its acquisition of privately held biotech Proteologix, adding bispecific antibody assets for atopic dermatitis to its pipeline. Proteologix’s PX128 is a bispecific antibody targeting interleukin 13 plus TSLP. The antibody is Phase I-ready for severe atopic dermatitis (AD) and moderate to severe asthma. Another preclinical antibody, PX130, targets IL-13 plus IL-22 for moderate to severe AD. Targeting dual pathways is expected to increase efficacy, according to J&J, and both candidates are designed for convenient, infrequent dosing intervals. ���Together, these pipeline additions demonstrate a strategic approach to build a portfolio of differentiated and complementary bispecifics,” J&J said in its announcement of the deal. In addition to the $850 million in cash, California-based Proteologix will also be eligible for an additional milestone payment. Our data shows that most patients with advanced NTRK fusion-positive solid tumors are not getting the right therapy, suggesting that targeted therapy for rarer and/or tissue agnostic alterations may get deprioritized or overlooked 2.
