"Under the physiological conditions, the probe WZL-1 exhibited significantly enhanced emission intensities at 630 nm for thiols cysteine, homocysteine, glutathione, and hydrogen sulfide. CAR T cell therapy targeting HER2 antigen shows promise against advanced sarcoma in phase I trial HOUSTON – (April 24, 2024) – Researchers at Texas Children’s Cancer Center and the Center for Cell and Gene Therapy at Baylor College of Medicine, Texas Children’s Hospital and Houston Methodist published results of a phase I clinical trial of a novel immunotherapy for high-risk sarcomas in the journal Nature Cancer. The therapy uses chimeric antigen receptor (CAR) T cells engineered to target the HER2 protein, which is overexpressed on the surface of sarcoma cells. The HEROS 2.0 trial showed that this therapeutic approach is safe and is associated with clinical benefit. “CAR T cell therapy has been a highly successful strategy for recurrent or high-risk leukemias or lymphomas, but challenges remain in using this therapy for solid tumors,” said first and corresponding author Dr. Meenakshi Hegde, associate professor of pediatrics – hematology and oncology at Baylor and pediatric oncologist at Texas Children’s Cancer Center. “The results of this trial show that we are moving the dial in harnessing the power of CAR T cells as an effective anticancer therapy for https://lnkd.in/eDn7Pdsv

Automated Mass Photometry of Adeno-Associated Virus Vectors from Crude Cell Extracts Mass photometry (MP) is a fast and simple analysis method for the determination of the proportions of subpopulations in an AAV sample. It is label-free and requires minimal sample volumes between 5–10 µL, which makes it a promising candidate over orthogonal techniques such as analytical ultracentrifugation (AUC), cryo-transmission electron microscopy (Cryo-TEM) or charge-detection mass spectrometry (CDMS). However, these methods are limited in their application to purified samples only. Here we developed a purification step based on single-domain monospecific antibody fragments immobilised on either a poly(styrene-divinylbenzene) resin or on magnetic beads prior to MP analysis that allows the quantification of empty, partially filled, full and overfull AAV vectors in crude cell extracts. This is aimed at identifying potentially promising harvest conditions that yield large numbers of filled AAV vectors during the early stages of the viral vector development platform, e.g., the type of transfection reagent used. Furthermore, we provide a direct comparison of the automated and manual handling of the mass photometer with respect to the quantities of AAV subspecies, molar mass of the capsid and payload, and highlight the differences between the “buffer-free” sample measurement and the “buffer-dilution” mode. In addition, we provide information on which candidates to use for calibration and demonstrate the limitations of the mass photometer with respect to the estimation of the capsid titer. https://lnkd.in/eRe7iG8T

RapidFire-MS was invented to address the limitations of traditional LCMS methods used in drug discovery and development. In the early 2000s, LCMS was the primary tool for analyzing large numbers of compounds in drug screening assays. However, it was relatively slow and oftentimes offered more separation than was necessary for MS analysis. To overcome long unnecessary chromatographic times, RapidFire-MS was developed as a high-throughput screening technology that could analyze large numbers of compounds in a short amount of time. The average cycle time for RapidFire was 8 seconds compared to many minutes for LC. This significant improvement in throughput was achieved by faster sampling, shorter flow paths, and bind/elute cleanup (instead of chromatographic separation). RapidFire-MS was designed to meet the demands of the pharmaceutical industry for faster and more efficient drug screening technologies. It has since become a widely used tool in drug discovery, metabolomics, and proteomics. In this study, we introduce a custom-designed lectin microarray featuring nine distinct lectins: rPhoSL, rOTH3, RCA120, rMan2, MAL_I, rPSL1a, PHAE, rMOA, and PHAL. These lectins have been specifically tailored to selectively bind to common N-glycan epitopes found in therapeutic IgG antibodies. By utilizing intact glycoprotein samples, our nine-lectin microarray provides a high-throughput platform for rapid glycan profiling, enabling comparative analysis of glycosylation patterns. Our results demonstrate the practical utility of this microarray in assessing glycosylation across various manufacturing batches or between biosimilar and innovator products. A careful look at lipid nanoparticle characterization: analysis of benchmark formulations for encapsulation of RNA cargo size gradient With the recent success of lipid nanoparticle (LNP) based SARS-CoV-2 mRNA vaccines, the potential for RNA therapeutics has gained widespread attention. LNPs are promising non-viral delivery vectors to protect and deliver delicate RNA therapeutics, which are ineffective and susceptible to degradation alone. While food and drug administration (FDA) approved formulations have shown significant promise, benchmark lipid formulations still require optimization and improvement. In addition, the translatability of these formulations for several different RNA cargo sizes has not been compared under the same conditions. Herein we analyze “gold standard” lipid formulations for encapsulation efficiency of various non-specific RNA cargo lengths representing antisense oligonucleotides (ASO), small interfering RNA (siRNA), RNA aptamers, and messenger RNA (mRNA), with lengths of 10 bases, 21 base pairs, 96 bases, 996 bases, and 1929 bases, respectively. We evaluate encapsulation efficiency as the percentage of input RNA encapsulated in the final LNP product (EEinput%), which shows discrepancy with the traditional calculation of encapsulation efficiency (EE%). EEinput% is shown to be < 50% for all formulations tested, when EE% is consistently > 85%. We also compared formulations for LNP size (Z-average) and polydispersity index (PDI). LNP size does not appear to be strongly influenced by cargo size, which is a counterintuitive finding. Thoughtful characterization of LNPs, in parallel with consideration of in vitro or in vivo behavior, will guide design and optimization for better understanding and improvement of future RNA therapeutics. https://lnkd.in/eQyPHJ7K

#TechnoteTuesday - Advancements in Plasmid DNA Analysis for Biopharma Plasmid DNA plays a crucial role in the biopharma industry, serving as a gene-delivery vehicle for DNA vaccination and as a key intermediate in processes like viral particle production for gene therapy and ex-vivo protein synthesis. Given the importance of plasmid DNA purity, federal regulations mandate a release criterion of >80% supercoiled content for manufactured injectable plasmid products. Traditional agarose gel methods have limitations in detecting low levels of impurities. To address this, our latest technical note explores the development of a rapid, sensitive, and automated method for plasmid DNA isoform analysis using Capillary Electrophoresis with Laser-Induced Fluorescence detection (CE-LIF). Key Benefits of Our Method: 1. Sensitive and Robust Method: A CE-LIF method for plasmid analysis was developed, providing baseline resolution of supercoiled, open-circular, and linear plasmid isoforms within 20 minutes. 2. Optimized Injection: Electrokinetic injection outperformed pressure injection due to more efficient injection of charged DNA analytes. 3. Enhanced Sensitivity: Utilizing SYBR Gold dye, known for its high quantum yield, results in higher sensitivity and a better separation profile through effective interaction with DNA. 4. Extended Capillary Lifetime: Incorporating a methanol rinse in the conditioning step extended the capillary's lifespan to over 100 injections without affecting topoisoform percentages. 5. Monitoring Plasmid Degradation: Experiments with stressed plasmid samples demonstrated that CE-LIF can accurately quantify plasmid topoisoforms, making it an excellent tool for monitoring plasmid degradation. This innovative approach offers a significant improvement over traditional methods, providing the biopharma industry with a more reliable means of ensuring plasmid DNA purity and integrity. However, obtaining FDA approval is only the first step.
