Addgene is a nonprofit plasmid repository located in Watertown, Massachusetts. Addgene’s mission is to accelerate research and discovery by improving access to useful research materials and information. Addgene facilitates the sharing of high-quality scientific materials, research reproducibility, and open science by archiving and distributing DNA-based research reagents and associated data to scientists worldwide. Addgene’s repository contains over 70,000 plasmids, including special collections on CRISPR, fluorescent proteins, chemogenetics, optogenetics, and viral plasmids. All plasmids in Addgene’s repository were deposited by your scientific colleagues from around the world. There is no cost for scientists to deposit plasmids, which saves time and money associated with shipping plasmids themselves. All plasmids are fully sequenced for validation and sequencing data is openly available. Addgene also provides ready-to-use AAV and lentiviral preparations of commonly requested plasmids as a service to scientists. Addgene offers free online educational resources and protocols about molecular biology topics such as common vector backbones, cloning, CRISPR, AAV and lentivirus production, and science careers. These resources include the Addgene blog, eBooks, guides, videos, and a podcast series.

💥 What are antibodies? Where are the epitopes and isotypes located? Affinity? Avidity? I stumbled over this great 12-minute video from Addgene, and it is an easy way to learn a lot more about our immune system, focussing on antibodies. 12 minutes on a Saturday morning perfectly well spent I guess. Enjoy! 🧬 An antibody is a Y-shaped protein produced by B cells that plays a crucial role in the immune response. Each antibody has a specific paratope that binds to a unique epitope on an antigen. The antibody structure includes two main regions: the Fab (fragment antigen-binding) region, which contains the paratope, and the Fc (fragment crystallizable) region, which interacts with immune cells. Antibodies are classified into five isotypes: IgG, IgA, IgM, IgE, and IgD, each with distinct functions. IgG is the most abundant, IgA protects mucosal areas, IgM is the first responder, IgE is involved in allergic reactions, and IgD's role is less defined. #immunesystem #cancervaccines

We compared the efficiency of base editors (APOBEC1, APOBEC3A, BEACON2, evoCDA1, ABE8e and ABE8.20-m) to induce splice site mutations and exon skipping before using them to cross-validate the exon hits. 4. We applied machine learning and statistical methods to identify the characteristics of exons that promote or suppress cell fitness. 5. We investigated the role of TAF5-exon8, a screen hit whose splicing is regulated by SRSF1, and found it to be crucial for TFIID complex assembly and gene expression.
