Be honest with your workload and what you can literally take on. I've found it helpful to list all my current tasks and then ask is that new task more important than any of the listed others. If not, negotiate a new time for that task like end of day tomorrow. That way you're looking after yourself (not overloading) and also giving your manager a realistic time frame for the task to be done setting expectations on both sides.
