Cationic carbon dots and their use to boost siRNA delivery: a matter of charge-based bond's strength. Today, we are going to talk about the use of cationic carbon dots (or CCDs) and their use for gene delivery. As nucleic acids are negatively charged, CCDs that contain abundant positively charged amine groups can bind to nucleic acids to form CCD–nucleic acid complexes and attach on the surface of the cell membrane to promote the endocytosis-based cellular uptake process. In a recent study, researchers have developed a type of CCD known as APCDs, using arginine (Arg) and pentaethylenehexamine (PEHA) as precursors, and conjugated them with oleic acid (OA) for gene delivery. By fine-tuning the mass ratio of APCDs to OA, 3x distinct APCDs-OA conjugates were synthesized: 1) APCDs-0.5OA; 2) APCDs-1.0OA; and 3) APCDs-1.5OA. These amphiphilic APCDs-OA conjugates exhibit high affinity to DNA through electrostatic interactions, with APCDs-0.5OA showing strong binding with siRNA. Some key highlights to be aware of: 🔬 Versatile distribution -upon internalization by HEK293 and osteosarcoma (U2OS) cells, these conjugates distribute in both the cytoplasm and the nucleus, which serve as active sites for siRNA and pDNA, respectively. ⚡ Enhanced GFP expression -the APCDs-OA conjugates successfully deliver GFP-encoding plasmid DNA into the cells. Notably, GFP expression levels mediated by APCDs-0.5OA and APCDs-1.0OA were 10x times higher than those achieved with PEI in HEK293 cells. 📈 Improved siRNA transfection -APCDs-0.5OA demonstrated significant siRNA transfection efficiency, effectively downregulating the expression of FANCA and FANCD2 proteins in U2OS cells to 9.9 and 19.1%, respectively. This study showcases how conjugation of CCDs with a lipid structure like OA can significantly enhance gene transfection efficiency. Pipeline of Antibacterial Agents: WHO assessment and recommendations Out of antibacterial 128 drug discovery programmes 16 made it to the market ( a success rate of 12.5 %). WHO has made recommendations which is worth noting. Interestingly , small biotech companies are doing major work in this area. Heartening to see SE asia contributing to this (eg Bugworks) One of the recommendations is to focus on non traditional therapies( again good to see Gangagen contributing to this area from SE Asia). We are extremely proud to share our article on leishmaniasis recently published with the team of Prof. Bhaskar Saha, National Centre for Cell Science, Pune University. Leishmaniasis is a complex parasitic disease that poses numerous public health challenges worldwide. In this article, we explore the fundamental mechanisms of the parasite, this understanding is necessary to find new valuable therapeutical strategies. This publication represents months of research, analysis, and collaboration with experts in the field. We trust this article will contribute to a better understanding of leishmaniasis mechanisms and encourage stronger initiatives to combat this disease without valuable therapeutic solutions. This work has already taken advantage of by our teams. Indeed, based among other things on this study, we are developing a new generation of mRNA vaccine against leishmaniasis. We combine the know-how of GEG Tech in vectorology and its unique biological mRNA platform with the deep knowledge of Prof. Bhaskar Saha teams and Ricardo Jorge Leal Silvestre teams (Minho University) about the leishmaniasis. Two animal studies have been led and completed, showing an unprecedented parasite protection for the vaccinated animals. We will let you know further advances of this new vaccine candidate. The below article talks about the benefits and disadvantages of breast cancer screening with abbreviated MRI. It finds 11.8 tumors whereas 3-D tomosynthesis finds 4.8 tumors. Abbreviated MRI appears to be the gold standard for finding breast cancer early. The downside is that there will be more false positives and more unnecessary biopsies. But that usually doesn’t result in death, unlike untimely breast cancer diagnosis. #breastcancerscreening #breastcancer

Learnings and Observations from MPS World Summit: 1. The MPS models are becoming more unique, complex and with greater interplay across human organ models 2. The ability to drive standardisation across various MPS platforms will be almost impossible 3. Standardisation should focus on the reagent’s (cell lines, cells, matrices and media) which means industrial scale, consistentancy batch to batch, reagent validation into the MPS platforms, stable and well characterised cell lines with appropriate rights and consents and finally quality systems alignment with the minimum standard being ISO9001, supported by adherence to ISSCR guidelines (if in the iPSC thematic) in the suppliers site of manufacture. 4. Good alignment and support being driven by regulators, government bodies and 3R promoting bodies to support market adoption and growth. Conference was well attend by FDA, NIH, 3R organisations and various research institutions with links to military/safety applications. 5. Biopharma interest, use and engagement continues to grow 6. Really engaging and positive MPS community has been established, this is leading to strong relationships and creates a collaborative culture to better enable science.
