At InsideTX, we are revolutionizing nanoparticle manufacturing, to unlock the full potential of RNA-LNP therapeutics and nanomedicines. At the convergence of microfluidics and biotechnology, we strive to provide laboratories, biotech, and pharma companies with the ultimate nanoparticle synthesis capabilities by solving three of the biggest challenges of the RNA-LNP revolution: Easing screeening, ensuring seamless scalability all along the development process and allowing for continuous production. In practices, our system allow for easy manufacturing of most lipidic or polymeric nanoparticles (liposomes, LNP, solid lipid nanoparticles, PLGAs…) using the same system all along your drug development process - from your screening stage at µL scale to your continuous production at 10s of L scale - while ensuring ultimate nanoparticle characteristics control at every step of the process.

    Embarking on the journey of Lipid Nanoparticle (#LNP) formulation for oligonucleotide/RNA delivery might initially appear complex and intimidating. However, rest assured, the process is more straightforward than it looks! Here's a straightforward guide to get you started:   1/ Identify Your Oligonucleotide/RNA 🧬: Whether it's #mRNA, #siRNA, sgRNA, or #ASO, the choice of nucleic acid hinges on your therapeutic goal.   2/ Lipid Selection 🔬: Numerous lipid combinations exist, each suited to different cargoes and cell/organ targets. We suggest starting with proven LNP compositions to ease the development process, before exploring more complex solutions. Find out more about those here: https://lnkd.in/eE7vA6UB   3/ Formulation Design 🧪: Picking the right buffers, pH, N:P ratio... is key. For example, lipids are dissolved in an organic solvent, typically ethanol. An acidic buffer is needed in the aqueous phase to ionize the lipids during complexation with RNA.    4/ Efficient Mixing Process 🌀: Employ a microfluidics-based system to mix aqueous and organic phases effectively and homogeneously, ensuring seamless RNA-LNP complex formation for optimal physicochemical parameters control.   5/ Parameter Optimization 🛠️: Fine-tune the synthesis conditions (flow rates and concentrations) to nail the perfect particle size, PDI, and encapsulation efficiency. Remember, these factors are key to efficient RNA-LNP intracellular delivery.   6/ Improve Selectivity (Optional) ➕ : Consider decorating LNPs with ligands or peptides for targeted delivery to specific cell types (also known as active targeting.)   7/ Test, Refine, Repeat 😎: Physicochemical characterization and, in vivo and in vitro testing are key. While general guidelines can be drawn, LNP formulation should be optimized for each payload and target.     Quelle quinzaine dense pour l'écosystème santé en Nouvelle Aquitaine ! La semaine passée se réunissait les membres du cluster santé ALLIS-NA | Alliance Innovation Santé Nouvelle-Aquitaine afin de renouveler son Conseil d'Administration auquel j'ai eu le privilège d'être élu. Sous la Présidence de Philippe Lamoureux, cette belle équipe, va œuvrer pour faire rayonner l'écosystème santé dans la Région Nouvelle-Aquitaine et au-delà. Ce vendredi dernier, nous participions à la Journée Nationale de la Bioproduction de Biothérapies, organisée par France BioLead (bravo Caroline Bernard !) et ALLIS NA (bravo Alice Leroux, PhD !). Je n'ose imaginer le travail de préparation derrière cette journée où une trentaine de sites de bioproduction ont ouvert leurs portes. Un immense merci de faire vivre notre filière et de nous permettre de nous rencontrer, de tisser des liens, de mailler le territoire, de pouvoir tout simplement échanger en passionnés sur les sujets santé ! L'occasion aussi de présenter notre projet Inside Tx de bioproduction de nano médicaments avec Thania. Merci à Michael et Frédéric pour leur accueil chaleureux et le récit de cette aventure extraordinaire qu'est TreeFrog Therapeutics : bravo et merci pour ce que vous faites.
